The American singer will be joined by Marina And The Diamonds, Juliette Lewis and Shakespeare's Sister for the "ultimate girls night out" at Friday's Big Top on June 11.
Further acts confirmed include Mini Viva, Woman, Daisy Dares You and I Blame Coco.
Florence and the Machine, N-Dubz and La Roux were recently added to the lineup for the three-day festival.
Isle of Wight takes place between June 11 and 13 at Seaclose Park, Newport.
