The singer will join the headlining rapper on the Sunday lineup, which also includes Mr Hudson and further acts to be announced in the coming weeks.
Jay-Z recently revealed that he was eager to collaborate with the 'Not Fair' star.
"I was very flattered to hear that Jay-Z wanted me to support him at Wireless and, combined with my dates in March with Dizzee Rascal, I am excited to end my current string of live shows in such prestigious company," Allen said.
The 24-year-old and Dizzee Rascal will co-headline two live dates at Manchester MEN arena on March 5 and London O2 arena on March 7.
Allen recently confirmed that she is temporarily quitting music to focus on opening a record label and shop.
