The duo will play the Roundhouse venue in Camden on July 17 with support from Bloc Party frontman Kele Okereke.
A statement on the band's website read: "Underworld return to active service with a one-off headline gig as part of the iTunes Festival at London’s Roundhouse in July.
"This show will be the first chance for people to hear the band previewing tracks from their forthcoming sixth studio album."
The dance group recently released a free download of new track 'Scribble', taken from the as-yet-untitled follow-up to 2007's Oblivion With Bells.
Other acts confirmed for this year's iTunes Festival include Diana Vickers, Kate Nash and Ozzy Osbourne.
