The ex-Beatle will support the Born HIV Free campaign with the first European concert broadcast live through the video-sharing website.
A statement on McCartney's website read: "Carla Bruni-Sarkozy and The Global Fund to Fight AIDS, Tuberculosis and Malaria are proud to announce that Paul is throwing his support behind the Born HIV Free campaign.
"His forthcoming show this weekend in London's Hyde Park will be broadcast exclusively through the Born HIV Free channel on YouTube."
The concert will remain on the site for a full month after its initial broadcast.
The supporting bill at Hard Rock Calling on Sunday includes Crosby, Stills and Nash, Crowded House and Elvis Costello among others
McCartney recently confirmed that the Manic Street Preachers will play at his upcoming gig at the Millennium Stadium on June 26.
