The group, whose fourth studio outing Only By The Night entered the album chart at number one in 2008, have sold over a quarter of a million digital copies of the LP.
Statistics compiled by the BPI and Official Charts Company reveal that the US rockers have sold 276,000 copies of the record online.
Lady GaGa's The Fame lands in second place with digital sales of 261,000.
According to The Daily Telegraph, managing director of the OCC Martin Talbot predicted that the record would soon be broken as the increase in legal downloads means that digital sales account for almost a fifth of album sales.
He said: "With Kings of Leon leading the way in the all-time list - Only By The Night becoming the first album to sell more than quarter of a million copies - it won't be long before we see the first half-million-selling digital album."
Kings of Leon recently unveiled details of their new album Come Around Sundown, slated for release on October 19.
