The stars, who are both nominated for awards at this year's ceremony, join Linkin Park, Katy Perry, Ke$ha, Kings of Leon and Kid Rock on the list of artists confirmed to appear.
Shakira is up for the 'Best Female' award against fellow performer Perry, Lady GaGa, Rihanna and Miley Cyrus.
Meanwhile, London rapper Plan B received nods in the 'Best New Act' and 'Best Video' categories.
In addition to the bill of performers, Desperate Housewives star Eva Longoria will host the evening, while The Dudesons, Emily Osment, Jackass's Johnny Knoxville and Jersey Shore's Pauly D will hand out awards.
The 2010 MTV Europe Music Awards will take place in Madrid on November 7.
