The Jamaican singer, who was best known in the UK for his 1982 album Night Nurse, passed away surrounded by his family on Monday morning, BBC News reports.
Isaacs was diagnosed with lung cancer last year, and had travelled around Jamaica before returning to London to spend time with his family and wife Linda.
Linda said in a statement: "Gregory was well-loved by everyone, his fans and his family, and he worked really hard to make sure he delivered the music they loved and enjoyed.
"He will be greatly missed by his family and friends."
Madness frontman Suggs, who Isaacs had supported on tour, paid tribute to the performer at this afternoon's Q Awards in London.
He told the audience: "He was a great reggae artist and also one of the most sartorially elegant stars on the world stage."
