The two pop bands recently confirmed that they will unite for a joint tour of the US next summer. They will also take to the stage together at the AMAs this month.
Speaking on Ryan Seacrest's KIIS-FM radio show, New Kids band member Donnie Wahlberg said: "I don't think we'll do a new song. I think we'll do a bunch of our hits together. I think the one thing about this tour is that it's creating a lot of curiosity.
"The beauty of it is that we have so many hit records between the two bands. We could go on stage for three hours and do fifty hit records. We want to do a taste of that on the AMAs and remind the people who don't know and show our fans what is in store."
Backstreet Boys star Howie Dorough added: "As of right now, we're putting together a little mash-up!"
In a statement, the show's producer Larry Klein added: "New Kids on the Block and Backstreet Boys on one stage is something special. Moments like these are what the AMAs are all about."
Other performers scheduled to take to the stage at the AMAs include Christina Aguilera, Justin Bieber, Katy Perry, Ke$ha and Usher, while Rihanna will open the show.
The 38th annual American Music Awards airs live from the Nokia Theatre in Los Angeles on November 21 from 8pm ET on ABC.
