Yolanda Adams, Martina McBride and Florence Welch will also take to the stage to honour The Queen Of Soul at the event in Los Angeles, it has been confirmed.
The performance will feature individual and group renditions of some of Franklin's best-loved tracks, while Esperanza Spalding is also scheduled to perform alongside members of the Grammy Jazz Ensembles and Grammy Camp.
It was previously speculated that Aguilera - who sang the national anthem at last weekend's Super Bowl - would open the ceremony with the tribute, although this has yet to be confirmed.
An official press release described the Grammys tribute as a "valentine and get-well card" for Franklin, who underwent surgery earlier this year.
It has also been confirmed that Marc Anthony, Jamie Foxx, Jewel, John Mayer, Pauley Perrette and Donnie Wahlberg have joined the star-studded lineup of presenters for the ceremony at the Staples Centre.
Other previously-announced performers include Justin Bieber, Rihanna and Drake, Eminem, Katy Perry, Lady GaGa and Bruno Mars, while the 'MusiCares Person of the Year' recipient Barbra Streisand will also perform.
The 53rd Grammy Awards airs Sunday, February 13 from 8pm ET/PT on CBS.
