The AP reports that the rockers will be joined by David Crosby and Graham Nash for the event, which is scheduled to take place next month.
The show will raise money for the Fund for Civility, Respect and Understanding, which was founded by shooting victim Ron Barber to aid those injured in the altercation.
Other acts included in the official line-up are guitarist Nils Lofgren, Sam Moore, Keb' Mo' and alternative country band Calexico among others.
Giffords's husband Mark Kelly is to appear onstage as well to give an address to the crowd.
The Fund for Civility, Respect and Understanding's benefit gig will be held on March 10 at Tucson's Convention Centre.
