McGarrigle, the mother of musicians Rufus and Martha Wainwright, died last year after battling cancer. The singer performed with her sister Anna in a Canadian folk duo.
The two-day event will take place at New York's Town Hall in May, reports Entertainment Weekly.
Rufus and Anna will perform at the concert alongside Jones and Fallon. Other artists including Emmylou Harris and Antony Hegarty are also scheduled to perform.
Funds raised at the event will go towards finding a cure for sarcoma.
