The band, who announced their reunion last week, are to use the slot to launch their comeback.
The 'I Believe In A Thing Called Love' stars split five years ago amid rows over frontman Justin Hawkins's drug addiction, which he has now recovered from.
Def Leppard headline the opening night on Friday, June 10 at Donington Park, Derby, and The Darkness will be special guests.
System of a Down and Linkin Park are to headline Saturday and Sunday respectively.
Tickets are on sale now for Download, which takes place from June 10 to June 12.
