The 'Higher' girl group will play the gig at London's Clapham Common on September 10, with tickets for the 2012 Paralympic Games going on sale on the same day.
The event, which is sponsored by Sainsbury's, will also include performances from Chipmunk and Taio Cruz.
Proceeds raised from the concert will go towards the Paralympic Legacy Fund.
Speaking about the group's involvement, band member Frankie Sandford explained: "This gig will turn the spotlight on the Games. We are really chuffed to be involved."
The Saturdays are currently preparing to release their new single 'Notorious' and recently shot the music video for the track in Los Angeles.
