The reorganised three-day event will take place at Paddock Wood, Kent between July 1 and 3, and will include previously-confirmed headline acts Morrissey and The Eagles.
"I am really excited about having Prince at the Hop Farm, we have worked hard to get the third day up and running and we hope that fans will appreciate the integrity of the bill and see it's one of the most musically exciting this year that stands head and shoulders above a lot of other festivals," said festival organiser Vince Power.
It will be the first UK performance from Prince since his record-breaking 21-date residency at London's O2 arena back in 2007.
Tickets for the third day will go on sale at 9am on May 19, with current two-day ticket holders able to upgrade their tickets for just £40.
Further acts confirmed to play the music event include The Human League, Bryan Ferry, Carl Barāt, Noisettes and the Guillemots, among many others.
Supporting acts for Sunday's lineup will be confirmed shortly.
