The pair performed the track, titled 'Rise Above 1', on the finale edition of American Idol on Wednesday night with the musical's star Reeve Carney.
U2 decided to release the track to iTunes in the US when it received a positive fan response.
Spider-Man: Turn Off The Dark has faced widely negative reviews and suffered a number of financial and technical issues before it was recently revamped.
U2 are currently finishing up their record-breaking world tour as well as working on a "futuristic" new album with Lady GaGa producer RedOne.
