The singer had been walking to the stage to hand out the 'Male Video of the Year' gong when she slipped on the walkway and crashed to the floor.
She immediately got up again and continued to the stage, where she presented Blake Shelton with the award for his 'Who Are You When I'm Not Looking' video.
"I've just made a complete fool of myself and experienced one of life's most embarrassing moments!" she later joked in a video posted on her official website.
However, the 'You're Still The One' star went on to laugh that the slip and her role in the action-packed Thelma & Louise skit that opened the show at least proved that she would never need a stunt double.
"I didn't hurt myself but I have a bit of a sore thumb," she added. "I'm going to auction off those shoes and get rid of them as soon as I can! I never want to see those shoes again... Or maybe I'll keep them as a souvenir..."
The singer starred alongside Taylor Swift in the Thelma & Louise sketch, which parodied the 1991 movie starring Susan Sarandon and Geena Davis. Donald Trump, Glee actor Chord Overstreet and ceremony host Kid Rock also made appearances.
Twain's appearance at the CMT Music Awards came following the announcement that she has agreed to a two-year residency at The Colosseum at Caesars Palace in Las Vegas beginning next year.
