The singer will close the music event on Sunday evening with a headline performance on the Pyramid Stage.
Beyoncé is thought to have originally asked Kelly Rowland and Michelle Williams to visit her at the festival, but has now suggested that the trio reunite on stage to perform a medley of their biggest hits.
"Originally the plan was simply for Kelly and Michelle to head down to Somerset to see their old bandmate," a source told The Mirror.
"Beyoncé was over the moon and sorted them out with Access All Area passes and accommodation. From there, plans developed and the idea of a brief on-stage Sunday evening reunion was mooted.
"It's still very much up in the air as no-one wants Bey to feel she's being upstaged. One idea is for the two girls to join her on stage towards the end of her set for a medley of their hits."
The source continued: "But whatever happens, the three girls are getting back together at Glasto and planning on having an absolute ball."
Destiny's Child disbanded in 2004, but rumours of a reunion have gained momentum over the past few months.
Reports have previously suggested that Beyoncé is planning a huge spectacle, as well as securing cameo appearances from Jay-Z and Kanye West.
