The US rapper watched Nutini from the side of the Pyramid Stage on Saturday afternoon and is said have been "excited" by what he saw.
"Jay-Z was getting really excited about Paolo. He'd heard about him before Glastonbury and was keen to see him in action," a source told The Sun.
"As soon as the show ended he approached Paolo and told him he was the best thing he'd seen so far at the festival. They talked about future projects - Jay-Z is keen to work with him."
The 'Empire State of Mind' hit-maker was at the festival to support his wife Beyoncé, who headlined with a 17-track set on Sunday night.
Jay-Z was also seen in the crowd for U2 and Coldplay's headline gigs, hanging out with fellow A-lister Gwyneth Paltrow.
