The former White Stripes frontman joined headliner Seasick Steve on stage at Camden's Roundhouse in London.
Alison Mosshart from The Kills and Led Zeppelin bassist John Paul Jones also took to the stage for the performance. Actor Jude Law was in the audience.
Seasick Steve, who reached number six on the UK album chart in June with You Can't Teach An Old Dog New Tricks, has recorded with White in the past.
Adele, Foo Fighters and Friendly Fires are all due to play the festival later this month.
