The band will headline the Apollo Stage on Sunday, July 10, with Limp Bizkit and Motorhead among the supporting acts.
The gig will be streamed on Slipknot's official website, as well as Sonisphere's site Sonispherefestivals.com.
A statement said that the show will be an "epic and emotional occasion", as it will mark their first UK gig since the death of bassist Paul Gray last year.
"England has always been a special place for this band from the very beginning. We can't think of a better way to culminate our celebration of Paul's life than to share it with the maggots at Knebworth and the millions more watching around the world. This is not to be missed," the statement read.
This will mark the first time in Sonisphere's history that a performance will be streamed.
The festival's opening night will include metal's 'big four' (Metallica, Slayer, Anthrax and Megadeth) take to the stage. Biffy Clyro and Weezer will headline the middle Saturday.
