The local borough council will decide on Tuesday, September 13 whether or not to approve the planning application from Oakmayne to build a residential block opposite the venue.
Writing on Twitter, Wretch 32 said: "Show some love and support for the Ministry. Stop them getting shut down."
Example added: "Please help save one of the World's most important clubs Ministry Of Sound from being closed down by signing here."
Armin Van Buuren tweeted the club to say: "Thank YOU for so many great memories. Thanks to people like you, guys like me can shine..."
Pete Tong said: "Ministry of Sound potentially under threat of closure, support them by signing the petition."

Over 22,000 people have currently signed the petition.
"Ministry of Sound is not just a nightclub," a statement from the club read. "We are the people that come here, the people that work here, the music that is played here and the lifelong bonds that are made here.
"If we lose Ministry of Sound, we are losing a vital piece of youth culture in this country. Nightclubs may have existed before Ministry of Sound, but they were never the same afterwards."
Last year, former Undertones frontman Fergal Sharkey expressed support for Ministry of Sound and Calvin Harris also signed the petition.
Ministry of Sound is currently celebrating its 20th year since it opened its doors on September 21, 1991. To mark the anniversary, its 20:20 Project is running throughout 2011 and culminates in a three-day party from September 16 to September 18.
A five-disc Live & Remastered compilation of sets from Larry Levan, David Morales, Kenny Carpenter, Todd Terry and Justin Berkmann will also be released on September 19.
