Sheeran tops the bill for the Manchester event, held on December 5 at the O2 Apollo, while Kaiser Chiefs headline the London event on December 14 at Brixton Academy.
Other live performances at the London show include The Maccabees, The Horrors, Miles Kane and Tribes, while Chase & Status, Nero, Fenech Soler and Maverick Sabre will play in Manchester.
Danny Wallace, London's Xfm Breakfast Show host, said: "Winter Wonderland is always awesome and this year's is set to be the most awesome yet. Add incredible bands to a venue packed with Britain's best radio listeners and you've got an amazing night to remember.
"I'm going to leave the venue halfway through, just so I can go twice."
Andy Ashton, Xfm's programme director, added: "Xfm's Winter Wonderland is a huge moment in the calendar for the station and its listeners. We pride ourselves on raising the bar with each year that goes by and this year is no exception."
The event, which is now in its eighth consecutive year, raises money in support of War Child. Tickets are available now on xfm.co.uk.
