The annual string of concerts will showcase the best of urban music across four cities, including Manchester, Birmingham, Bristol and London.
Rowland will headline the final night of the tour at London's Brixton Academy on December 1, along with Jessie J, Big Sean and the UK Female Allstar MCs, which is a one-off collaboration with Mz Bratt, Lady Leshurr, RoxXxan, Liones, Baby Blue and A.Dot.
Speaking about her participation, Rowland said: "I loved performing at the first ever 1Xtra Live four years ago and I'm really happy to be headlining the final date of this incredible event."
She added: "There's so much amazing talent on the lineup and I'm proud to be part of it."
Meanwhile, Wretch 32, Dappy, Labrinth and Tinchy Stryder will play the opening night at the Manchester Apollo on November 28.
Nero will headline the show at Birmingham Academy on November 29 and will be preceded by Skream, J. Cole and Maverick Sabre.
Chase & Status will headline the Bristol Academy date on November 30, while Benga and Youngman, Ms Dynamite and Delilah also appear on the bill.
MistaJam will co-host all four shows and will be joined by Twin B, Gemma Cairney and Trevor Nelson at select dates.

Passes will be limited to two per person and registration to be included in the ballot will close at 9am on November 11.
