The Florence Welch-fronted band will play six tracks at the Maida Vale studio on November 25, which will be broadcast live on BBC Radio 1, online and via the Red Button.
Fearne Cotton will host the show, which will include tracks from the group's new album Ceremonials as well as a special cover version.
Speaking about the special gig, Cotton said: "I'm really looking forward to hearing Florence + the Machine in the Live Lounge again.
"It's going to be amazing, especially as this is an extended set. All our Live Lounge acts are fantastic but I bet Florence has something special up her floaty sleeve for our lucky live audience and I can't wait to hear it!"
Florence + the Machine topped the UK chart with Ceremonials earlier this month.
Adele was the first artist to perform an extended Live Lounge session back in February 2011, where she covered Cheryl Cole's 'Promise This'.
Cee Lo Green played an extended session back in September, while Florence + the Machine will be just the third act to do so.
