The London-born rapper joins the bill of the Seaclose Park event along with Labrinth, Band of Skulls and Joan Armatrading.
Tempah was scheduled to appear at the 2011 Isle of Wight Festival before pulling out. The artist completed a UK arena tour earlier this year.
Bruce Springsteen & The E Street Band, Pearl Jam and Tom Petty and the Heartbreakers will headline next year's event. Groove Armada will present Red Light to perform a dance set in the Big Top tent, while Crystal Castles will headline the The Garden Stage on the Friday.
Other acts previously confirmed for next year's Isle of Wight festival include Noel Gallagher's High Flying Birds, Biffy Clyro, Example, Madness, The Vaccines and Noah & The Whale.
The Isle of Wight Festival takes place at Seaclose Park between June 21 and June 24, 2012.
A full lineup for the event is to be announced in the New Year.
