The musician, who has featured on both of the group's studio albums, confirmed the news on his official website, but failed to give a reason for his departure.
Tillman said: "Farewell Fleet fans and friends. Back into the gaping maw of obscurity I go. Tokyo is my last show with the Foxes. Sorry if I was distant and obtuse if we ever met. Have fun."
The 30-year-old has released seven solo albums and three EPs since 2005 under the name Father John Misty.
Fleet Foxes have yet to confirm a replacement or release a statement about Tillman's departure.
The band are nominated for best 'International Group' at next month's Brit Awards and face competition from Foo Fighters, Jay-Z and Kanye West, Lady Antebellum and Maroon 5.
