The singer will make her live performance comeback at the ceremony, which will be held at the Staples Centre in Los Angeles on February 12.
Announcing the news, Adele tweeted: "Ima be, Ima be singing at the Grammys. It's been so long I started to forget I was a singer! I can't wait, speak soon xx (sic)."
She later said in a statement: "I'm immensely proud to have been asked to perform at this year's Grammy Awards.
"It's an absolute honour to be included in such a night, and for it to be my first performance in months is very exciting and of course nerve-racking, but what a way to get back into it all."
The star has been unable to sing live over the past few months, after undergoing emergency microsurgery on her throat to stop recurrent vocal cord haemorrhage from a benign polyp.
Adele is nominated for six awards at the Grammys 2012, including 'Album of the Year' and 'Best Pop Vocal Album' for 21 and 'Song of the Year' for 'Rolling In The Deep'.
Paul McCartney, Coldplay, Rihanna, Nicki Minaj, Taylor Swift, Foo Fighters and Kelly Clarkson are among the artists previously confirmed to perform at next month's show.
