Singers such as Sir Elton John, Sir Paul McCartney, Sir Cliff Richard, Dame Shirley Bassey, Sir Tom Jones, Gary Barlow, Cheryl Cole, will.i.am and Robbie Williams took to the stage.
Tom Jones, who performed hits such as 'Delilah', told CBS News that it was a "once-in-a-lifetime" opportunity.
"When you get a royal wedding or a Jubilee - and it brings people together, you see happiness on people's faces... It's very exciting," he said.
The Queen's husband Prince Philip was taken to hospital suffering from a bladder infection earlier in the day, forcing him to miss the concert.
"I hope he's okay," said Sir Paul McCartney, who closed the concert. "We all send our best wishes for a speedy recovery."
