The event was launched in 2009 to mark the 250th anniversary of Arthur Guinness signing the 9.000 lease on the famous Dublin brewery.
On September 28, Jones will headline the event at London's Senate House, with support from Brown, Wretch 23 and others.
Jones will also visit one of the 28 official Arthur's Day pubs in the vicinity of Senate House to perform an additional intimate set, which will also be broadcast on YouTube.
"I have great memories from playing the first Arthur's Day back in 2009, "Jones said. "Bringing it to Great Britain for the first time is going to be just as exciting.
"I'm ready to rock London, like we rocked Dublin in the first year. People can tune into YouTube on the night where they can watch my performance live. I hope everyone's ready to paint the town black!"
VV Brown added: "I've heard so much about Arthur's Day and to be amongst the first people to bring it to Great Britain is a real honour, especially with such a great lineup both in GB and globally.
"The night is going to be an eclectic mix of music, so there is something for everyone on
September 27. See you there!"
Wretch 32 said: "I can't wait to celebrate Arthur's Day with the British public and kick off the first event in GB.
"I'm really looking forward to performing alongside Mr Tom Jones and a raising pint of the black stuff to Arthur!"
Example, Tinie Tempah, Ellie Goulding, Mika, Professor Green, Fat Boy Slim, Texas and Amy McDonald will join the global celebration in over 55 countries.
