The Oxford rockers will perform a series of songs from their In Rainbows LP at the Rough Trade East shop at 8pm.
Entrance is on a first come first served basis, with doors opening at 7pm.
Guitarist Jonny Greenwood revealed on the band's Dead Air Space site that fans queueing for the gig will be given numbers so that they can get "snacks" and go for "toilet breaks".
He added: "For those who can't get in - and it's pretty small in there - we'll have some screens and speakers outside, if we're allowed. I think we are. And we'll also webcast it."
A link for the webcast will be on the band's official site later today.
