The Brighton DJ will take to the stage at London's Hyde Park on Saturday, July 5.
"For a long while I've wanted to play outdoors in London, but I just couldn't find a beach!" said the DJ, real name Norman Cook. "Hopefully we can bring some of the spirit of [my] Brighton shows into the heart of London town."
Electronic duo Underworld and funk icon Bootsy Collins - making his first UK live appearance in seven years - will also appear on the Saturday bill.
Other headliners at the four-day music extravaganza, which takes place on July 3-6, will be announced in the coming weeks.
Kaiser Chiefs, Klaxons and Basement Jaxx have performed at the event in previous years.
