The duo will sing together at John's AIDS Foundation Academy Awards to help raise funds for the charity.
John founded the charity in 1992 and has raised $125 million in 16 years. It has not yet been revealed what song they will play.
In a statement, Blige said: "Elton John is one of the most inspirational musicians and philanthropists in this industry for me.
"I respect him with all my heart, and it is truly a pleasure to be performing with him on such an important night as we look to raise awareness and vital funds for the fight against HIV/AIDS."
Blige and John have teamed up before on a rendition of 'I Guess That's Why They Call It The Blues' that featured on the albums Mary J Blige & Friends and Elton John One Night Only - The Greatest Hits.
