Weller will perform with Ocean Colour Scene guitarist Steve Craddock on April 10. Current chart-topper Duffy is also on the bill for that date.
The Mighty Boosh's Noel Fielding will host a comedy night on April 9, while the Fratellis and Figure 5 perform on April 11.
Matt Bellamy stated earlier in the week that his band Muse would play the event, with their gig now being confirmed for April 12.
The Teenage Cancer Trust show, co-organised by The Who's Roger Daltrey, has previously featured artists such as Noel Gallagher, Kaiser Chiefs and Razorlight.
