Damon Albarn's group will be joined by acts including Jerry Dammers from The Specials, Hard-Fi, Jay Sean, Patrick Wolf, Roll Deep and The Paddingtons.
The show, which celebrates the 30th anniversary of the Rock Against Racism concerts, takes place at London's Victoria Park on Sunday, April 27.
The 1978 event featured a concert from The Clash and saw 100,000 protesters take to the streets of London.
Due to Pete Doherty's recent arrest, Babyshambles will be replaced by the group Helsinki. They will feature guest members such as former Arctic Monkeys star Andy Nicholson, Reverend and the Makers singer Jon McClure and various members of Babyshambles.
Speaking about the show, Babyshambles bassist Drew McConnell said: "This has got to be the most important event of the year.
"Dozens of the best acts around, a huge celebration of multicultural London, and a chance to help stop those BNP racists gaining a foothold in the capital."
