The pop queen, currently number one on the UK singles chart with '4 Minutes', will take to the stage at Maidstone's Mote Park on May 10 - the first of the event's two days.
Fellow female soloists Duffy and Robyn will also perform that day, with The Feeling, The Hoosiers and Usher playing sets as well.
The event's second-day lineup includes sets from The Kooks, Adele and Goldfrapp, as well as a rare UK performance from US hip-hop star Nelly.
Radio 1's Big Weekend takes place at Mote Park, Maidstone, Kent on May 10 and 11.
