The singer said he was confident that Jay-Z would prove a hit at the summer bash, despite Gallagher's claims that hip-hop was "wrong" for the festival.
Usher told The Sun: "It really shocked me that he said that. Jay-Z is an incredible artist and headliner.
"I saw him perform at the Hollywood Bowl in LA, and you’re not getting better than that."
Glastonbury boss Michael Eavis hit back at Noel Gallagher's criticism last week, describing the Oasis guitarist's remarks as "weird" and "disappointing".
Jay-Z, Kings of Leon and The Verve headline this year's Glastonbury on June 27-29.
