The troubled singer will play the Pyramid Stage at the summer festival on June 28. Her performance will be followed by US rap star Jay-Z.
Eavis claimed she isn't worried about the possibility of Winehouse not turning up to the gig.
"I have total faith, I think she'll come and deliver," she told the BBC.
"This is one of the biggest sets of the summer, supporting the most anticipated artist at Glastonbury in the past 38 years."
Commenting on tabloid speculation about the soul musician's health, Eavis added: "I don't know how much is hot air from the press and how much is reality but she is pretty reliable actually.
"It's her personal life and no-one really knows the extent of what she's up to and I don't think it should be public knowledge.
"We all know she's a brilliant performer and I think that's what we should be focusing on."
Glastonbury takes place at Worthy Farm, Pilton from June 27 to 29. Tickets are still available.
