The pair competed at the same stage last year in a match that lasted 11 hours 5 minutes - the longest in tennis history. The match, which saw several other records broken, had to be played over three days.
On that occasion, Isner beat Mahut 6-4 3-6 6-7(7-9) 7-6(7-3) 70-68.
British number one Andy Murray tweeted his reaction to the draw, posting: "Isner vs mahut drawing each other in the first round after last year is the most amazing thing I've seen in tennis! Centre court anyone?!"
Meanwhile, American star Andy Roddick said on his Twitter account: "Isner/ mahut again. Unreal Do they play it on the same court???"
Doubles specialist Bob Bryan added: "Isner & Mahut! First round! Again! Gimme a break!"
Since the marathon match, Isner has revealed that he and the Frenchman have become friends.
He said: "Honestly, before Wimbledon, we'd never said one word to each other. Now we text each other quite often and I've got to know that he's a really class act and a really good guy."
