At 2pm at the Sensation stage in the Theatre field, the 9001st spot was laid. The players moved across the mat as the compere called out instructions from the stage.
The event also drew hundreds of spectators who stood around the edges of the mat and applauded when the world record was broken.
Instructions included moves that had to cross other people, as well as the standard combination of colours and limbs.
After ten minutes, so few people had fallen over that the compere announced that everyone still in the game was a winner.
The Twister games continued for another few minutes, before stickers were given out to players.
Spectators were then handed Twister spinners so they could continue playing.
Ms Dynamite plays the Wow! stage tonight in Glastonbury's Dance Village between 11.15pm and midnight. U2, Coldplay and Beyoncé will be headlining the festival's Pyramid Stage during the weekend.
