A spokesperson for the Charlie's Angels actress told People: "I can confirm the split with no additional comment."
The pair became a couple last summer after meeting on the set of the comedy film He's Just Not That Into You.
Long had earlier described Barrymore as "the most compassionate person I've ever met", while the actress said their relationship was "healthy, productive, supportive and full of humour".
They had reportedly been making plans to marry in the Caribbean.
