Dreyfuss has claimed that he gave $870,000 (£453,000) to his relatives in 1984 when they owned an interest in a Los Angeles office building.
In the suit, which was filed on Friday, Dreyfuss said that the loan is still unpaid and his uncle, Gilbert, has repeatedly declined to turn over financial records.
The 60-year-old actor is seeking repayment of the loan from his father Norman and uncle, plus interest and punitive damages.
Dreyfuss, an Oscar winner for 1977's The Goodbye Girl, has also starred in Close Encounters Of The Third Kind, Stand By Me and Always. He will next be seen in W. portraying vice president Dick Cheney.
