The presenter will play nightclub act 'Zaza' in the musical La Cage Aux Folles from January 19.
Norton attended a press conference at the Playhouse Theatre in full drag to make the announcement this morning. He is scheduled to appear until April 25.
The musical, which tells the story of a gay couple living in St. Tropez, was originally written as a play in 1973.
It became Broadway's first gay-themed musical when it opened in 1983, later going on to win six Tony Awards.
Norton will take over the role from actor Douglas Hodge. His first performance will be at the Royal Variety Show on December 11.
