The 60-year-old actor, who initially filed suit in August, has alleged that the pair borrowed $870,000 (£562,000) from him in 1984 in order to save a Los Angeles office building they owned.
Dreyfuss has said that the loan remains unpaid and that his uncle, Gilbert, has repeatedly refused to repay the money owed.
Last month in court documents, Gilbert Dreyfuss admitted that he had made some financial mistakes in the past but that payments on the loan were current. He also denied that the loan was as big as his nephew claimed.
The Oscar-winning actor, who recently appeared in W., has now amended his original lawsuit, claiming that his relatives have indulged in further financial wrongdoing.
Dreyfuss has alleged that the pair diverted millions of dollars they won from a previous legal battle and have failed to repay a $13.6 million (£8.6 million) loan on the office building, meaning foreclosure proceedings have begun.
