The former Sopranos star will appear alongside Marcia Gay Harden and Jeff Daniels in the comedy about two married couples who meet to talk after their children get into a playground brawl.
The role will mark Gandolfini's first return to Broadway in 14 years. He made his debut in a production of A Streetcar Named Desire in 1992.
Carnage received critical acclaim when it was staged in London last year with Ralph Fiennes starring.
The production is scheduled to open on March 22, with previews beginning February 28.
