The singer, who split from Jennifer Aniston earlier this month, had previously denied Hilton's claims that the pair made out for five minutes at a club in New York.
Speaking during a stand-up performance on the Mayercraft Carrier 2 Cruise, he said: "He [had to go] tell people that I made out with him at a club. I never said anything but you know what, you're damn right I made out with him at a club."
Last year, Hilton told Ryan Seacrest's KIIS-FM radio show that Mayer had "kissed me and I kissed him back - it was on the mouth with tongue".
The blogger, who offered to take a lie detector test to prove his story, added: "I thought he was messing with me. Then he kept going and going."
