Hilton asked Prejean during the Miss USA contest if she thought every state should follow the recent example of Vermont, which became the fourth in the union to legalise gay marriage.
Prejean took an anti same-sex marriage stance, answering: "In my country, in my family, I think that I believe that a marriage should be between a man and a woman. No offence to anybody out there, but that's how I was raised and that's how I think it should be - between a man and a woman."
The celebrity blogger was angered by the beauty queen's response and later posted a video blog, calling her a "dumb b*tch".
Socialite Kardashian, 28, has backed Hilton, describing Prejean as "narrow-minded".
"I'm not so narrow-minded so I definitely think a lot broader," she told People. "Everyone has the right to be happy and be treated equally and I think not allowing gay marriage just kind of puts us back.
"I believe you're in love with who you're in love with, and you should be able to marry them. No one should tell someone else how to think or how to feel."
She added: "I obviously have a completely different opinion but I think her answer was true to her. That answer represented her and how she felt. Everyone is entitled to their own opinion.
"She stood up for what she believes in so she should be happy with that backlash."
