The billionaire donned fake tattoos, greasy hair and mouldy teeth for an advert to promote the company's London-Scotland route.
Speaking about the new campaign, Branson told The Mirror: "I was never brave enough to come home with a tattoo when I was younger, so it's great to finally rebel.
"But I'm so proud of all our staff. And if dressing up in dirty clothes, having my teeth blacked out and getting covered in tattoos was what it took to prove it, I was delighted to oblige."
The commercial was filmed in Glasgow Central Station and airs from tonight.
