The 51-year-old hosted amfAR's annual Cinema Against AIDS function, where she remembered Richardson's contributions to the organisation, People reports.
"This year we lost an angel from our amfAR family. It was one of those random events that leaves you in a state of shock and profound grief," Stone said of Richardson's death.
She continued: "People say, 'Oh, she can't really have been that great or graceful'. She was that funny, charming, sweet and elegant. She was. She left behind her husband and two stunningly beautiful children and an unfinished mission to help us find a cure for AIDS."
The Casino actress also read a note from Richardson's husband, actor Liam Neeson, urging everyone to continue the charity's "great work".
Richardson died in March from a head injury sustained in a skiing accident, aged 45.
