The dermatologist said during an interview on Larry King Live that his contribution was "absolutely not" intended for Jackson and his then-wife Debbie Rowe, who worked for the medical practitioner, People reports.
However, Klein said that he could not answer for the paternity of Prince Michael, 12, and 11-year-old Paris Katherine, after it was speculated that neither Jackson or Rowe were their biological parents.
He said: "I still can't answer absolutely one way or another. I once donated sperm."
Klein insisted that the most important thing is "who the children want their father to be" and that the King Of Pop was a doting dad.
"Michael loved those children as a father. Those children loved him as a father. Because they would never pass him without saying, 'I love you, daddy'. He would say, 'I love you'. I've never seen such emotional care."
He added that he would be subject to a DNA test but does not want to see the familial bond the kids had with their parent tarnished.
"I don't want to destroy this relationship in any way, shape or form. I'll tell you this, no matter what, I will protect these children," he said.
Klein had previously denied that he was the father of the children, while he also refuted claims that he may have given the pop superstar illegal drug prescriptions.
