The 56-year-old actor will reportedly appear as his child-like alter ego for The Pee-wee Herman Show in Los Angeles in November.
According to the Los Angeles Times, the performance is a reimagining of his 1980s theatrical production that was also a special for HBO in '81 and led to his '85 feature film debut Pee-wee's Big Adventure. The franchise also includes children's programme Pee-wee's Playhouse, which aired on CBS from 1986 to 1990.
"I've put part of him away for a long time, but part of him has always been here with me," he said. "I think it will be like riding a bike - which is not a bad analogy for Pee-wee, by the way."
Reubens added that he will incorporate popular characters from his Saturday morning TV series into the show.
He added: "It has a lot of new material and a lot of old material. I felt that doing this show so many years later and having the TV show in between - people are going to ask, 'Where's the talking chair?'
"So I've added characters from the TV series to the show."
The Pee-wee Herman Show will run from November 19 to November 29 at the Music Box @ Fonda in Hollywood.
It was reported earlier that actor Johnny Depp was approached about taking over the role for a series of new films.
